THP1-XBlue™-defMyD Cells
THP1-XBlue™-defMyD cells are THP1-XBlue™ cells deficient in MyD88 activity. MyD88 is an adaptor protein involved in many signaling pathways. THP1-XBlue™-defMyD cells are unable to respond to the activation of receptors which signal through MyD88, such as TLR2, TLR4 and IL-1Rs. They remain responsive to MyD88-independent receptors, such as NOD1 and TNFR.
THP1-XBlue™-defMyD cells express an NF-κB/AP-1-inducible SEAP (secreted embryonic alkaline phosphatase) reporter gene. Activation of MyD88-independent signaling pathways in THP1-XBlue™-defMyD cells triggers the activation of NF-κB and AP-1 and the subsequent production of SEAP. The reporter protein is easily detectable and measurable when using QUANTI-Blue™, a medium that turns purple/blue in the presence of SEAP.
