THP1-defASC cells
To determine if a given signal is an inflammasome inducer
THP1-defASC cells are derived from THP-1 human monocytic cells. THP-1 cells are the most commonly used model cell line for the study of inflammasome activation as they express high levels of NLRP3, ASC and pro-caspase-1.
THP1-defASC cells express negligible levels of ASC but express native levels of NLRP3 and pro-caspase-1. These cells are unable to respond to inducers of ASC dependent inflammasomes, such as inducers of the NLRP3 inflammasome. THP1-defASC cells are designed as tools to determine whether a signal activates the inflammasome and in particular the NLRP3 inflammasome.
The positive control cell line for inflammasome studies are THP1-Null cells.

Growth Medium: RPMI 1640 (2 mM L-glutamine, 1.5 g/L sodium bicarbonate, 4.5 g/L glucose, 10 mM HEPES and 1.0 mM sodium pyruvate) with 10% heat-inactivated fetal bovine serum, 100 μg/ml Normocin™, Pen-Strep (50 U/ml-50 μg/ml)
