RNase L Activator
A cell-permeable tricyclic fused phenolic compound that induces RNase L dimerization and activation (EC50 = 22 µM) via direct interaction (KD = 12 µM) at the natural activator 2-5A (5'-phosphorylated, 2',5'-oligoadenylates) binding domain. Shown to exhibit broad-spectrum antiviral activity against diverse types of RNA viruses in both murine (EMCV & VSV in MEF) and human (HPIV-3, Sandai, XMRV in HeLa M, HeLa S, and DU145, respectively) cultures, but not in RNase L-deficient MEF culture or HeLa cells expressing inactive RNase L. Although the synthetic compound binds RNase L with a 4- to 6-times slower kinetic and is 44,000-times less potent than a trimer 2-5A (p3A2'p5'A2'p5'A), it is much more permeant and displays no cytotoxicity to MEF or HeLa S cells (50 µM for 24 h).
