Application Notes:
Nuclear Extract (source of HAT enzyme activity): Prepare nuclear extracts using a modified protocol of Dignam, et al., Nucleic Acids Res. 22: 1475, 1983 (recommended, but other methods may be used). See citations for reference link. Use 1- 20μl per assay point in the HAT Reaction Cocktail. Optimal nuclear extract concentration will need to be determined empirically.
