Conditions for 100% Activity
[1X Buffer G] + SAM:
[10 mM Tris-HCl (pH 7.5 at 37°C), 10 mM MgCl2, 50 mM NaCl and 0.1 mg/ml BSA] + 0.01 mM S-adenosylmethionine.
Incubate at 37°C.
Storage Buffer
Eco57I is supplied in:
10 mM potassium phosphate (pH 7.4 at 25°C), 100 mM NaCl, 1 mM EDTA, 1 mM DTT and 50% (v/v) glycerol.
Ligation and Recleavage
After 10-fold overdigestion with Eco57I, approximately 70% of the DNA fragments can be ligated, but none of these can be recut due to the methylation of the recognition sequence by this enzyme. |
